The mechanism of Krüppel-like factor 17 inhibit migration and invasion ability of renal cell carcinoma 786-0 cells

Objective To investigate whether Kruppel-like factor 17 (KLF17) could inhibit the migration and invasion ability of 786-0 cells by regulating the epithelial-mesenchymal transition (EMT). Methods Design and synthesize the specific small interfering RNA (siRNA) that can down-regulate the expression of KLF17 in 786-0 cells. SiRNAs were transfected into 786-0 cells and the effect was detected by Western blotting and real-time quantitative polymerase chain reaction (Real-time PCR). Migration and invasion ability were tested by the wound scratch assay and the Transwell chamber assay. The expression levels of EMT related molecules β-catenin, E-cadherin and N-cadherin were detected by Western blotting. Results SiRNA can effectively down-regulate the expression of KLF17 in 786-0 cells (P<0.01). Down-regulation of KLF17 expression can improve the cell migration and invasion capacity. The cell number in negative control group, siRNA1 group, siRNA2 group was 18.16±2.21, 29.03±2.48 and 28.93±3.00 respectively (P<0.01). In addition, the expressions of E-cadherin and β-catenin were decreased, while the expression of N-cadherin was increased in siRNA groups. Conclusion KLF17 may regulate EMT to inhibit migration and invasion in 786-0 cells. KLF17 may be associated with the development and metastatic in renal cell carcinoma.
