Detection of quaternary ammonium compounds-resistant gene and sulfamethoxazole/trimethoprim-resistant gene in Klebsiella pneumoniae.

OBJECTIVE To investigate the presence of the quaternary ammonium compounds-resistant gene and sulfamethoxazole/trimethoprim-resistant genes in Klebsiella pneumoniae isolated from the PLA 98th Hospital,Huzhou,Zhejiang province.METHODS Twenty-five strains of K.pneumoniae were isolated from the inpatients from Sep 2005 to Apr 2006.Five kinds of genes of qacE△1-sul1,dfrA1,dfrA5,dfrA12 and dfrA17 were analyzed by PCR and verified by DNA sequencing.RESULTS In 25 strains of K.pneumoniae,the positive rate of genes of qacE△1-sul1,dfrA5 and dfrA12 were 84.0%,24.0%,and 24.0%,respectively.Both genes of dfrA1 and dfrA17 were tested negative.The total positive rate of 5 kinds of genes was 92.0%(23/25).qacE△1-sul1 Positive gene sequencing showed highly homology with the sequence of GenBank(GenBank Accession Number:U12338).CONCLUSIONS There is very high positive percentage of qacE△1-sul1 gene in K.pneumoniae isolated from the inpatients.There is a close relation of the sulfamethoxazole/trimethoprim resistance in K.pneumoniae to qacE△1-sul1,dfrA5 and dfrA12 existing.
