Evaluation of three kinds of molecular diagnostic kits for Vibrio parahaemolyticus detection

Objective To evaluate the efficiencies of three kinds of molecular diagnostic kits for Vibrio parahaemolyticus(VP) detection.Methods The three kinds of commercial kits were Vibrio parahaemolyticus PCR kit from TIANDA company,Real-time PCR Kit from TAITAI GEN company and LAMP kit from HF company.A series of 10-fold dilution of pure VP enrichment broth were used to determine the limits of detection(LOD) of each kit and then sensitivity,specificity and agreement rate were compared with the National standard method(GB/T 4789.7-2008).Besides,the whole processing time,the costs and the simplicity of operation of different kits were also compared.Results The LOD of Vibiro parahaemolyticus PCR kit,real-time PCR kit and LAMP kit were 1.18×103 cfu/ml,11.8 cfu/ml and 11.8 cfu/ml,respectively.The sensitivity of all three kinds of kits were all 100% and the specificities were respectively 56%,48% and 50% when detecting for the seafood.There was no statistically significant difference among the three kits(P=0.074).The processing time of the PCR kit,Real-time PCR kit and LAMP kit were 120 min,80 min and 80 min,respectively.The costs of the kits were 10 Yuan,40 Yuan and 45 Yuan,respectively for a single sample.The LAMP kit was the simplest to operate,and the PCR kit was the most difficult one.Conclusion Three kinds of commercial kits were all have high sensitivity and low specificity,and all of them were easy to generate false-positive results.So they were recommended to be the screening tests for the detection of Vibrio parahaemolyticus.
