A Fluorescence Polarimetric Method for the Determination of Thrombin Based on Its Interaction with Aptamer

The anti2thrombin aptamer can interact with thrombin in high affinity. This interaction results in the change of the fluorescent polarization in the solution of the aptamer labeled with fluorescence. We developed a new method based on the change of the fluorescent polarization resulted from this interaction to detect thrombin. The linear range of the determination is 0. 1~4 IU/ mL . The detection limit for thrombin is 0. 09 IU/ mL . This method has been used to determine thrombin in human whole blood. The results obtained are satisfactory.
