Mechanism of sodium salicylate on paraquat-induced effects of NF-κB, TNF-α, and TGF-β1 in rat lung.

[Objective] To investigate the potential protective mechanism of sodium salicylate(NaSAL)on the pulmonary damage induced by paraquat(PQ)in rats.[Methods] We established rat poisoning model by intragastric administration of 80mg/kg PQ.A total of 72 male SD rats were randomly divided into control group,PQ treated group and PQ plus NaSAL intervention group.Control group were orally administered with an injection of 0.9% NaCl at the volume of 0.005 mL/g of body weight;PQ treated group were made 80 mg/kg PQ orally;PQ plus NaSAL group were given an intraperitoneal injection of 120 mg/kg NaSAL after an orally administration of 80 mg/kg PQ.Animals were sacrificed in batches on days 1,3,7,14 after experiment.Then,the level of nuclear factor of kappa B(NF-κB)was assayed by electrophoresis mobility shift assay(EMSA),while the protein levels of tumor necrosis factor-α(TNF-α),transformin growth factor-β1(TGF-β1)were determined by enzyme-linked immunosorbent assay(ELISA).[Results] After 80 mg/kg PQ exposure,the levels of TGF-β1,TNF-α protein expression and the rat lung tissue NF-κB activity increased more remarkably than that of the control group(P 0.05).In comparison with PQ treated group,with the treatment of NaSAL(120 mg/kg),the level of TNF-α,TGF-β1 protein expression and the rat lung tissue NF-κB activity decreased significantly(P 0.05)and trended to normal values.[Conclusion] Treatment of NaSAL could decrease the level of TNF-α,TGF-β1 protein expression and the rat lung tissue NF-κB activity.It suggested that NaSAL could alleviate paraquat-induced pulmonary damage in rats to some extent.
