Microscopic Observation of Cell Damage on Poplars Spears by Microdialysis Probe

When we use microdialysis technique to study the composition of plant sap,the primary problem we encountered is whether the dialysis fluid came from the apoplast or symplast? In this study,we used seedlings of winter hydroponic Populus.wutunenses and holland young as the material,and made the slice of cross-section tissue of spears damaged part after micro-dialysis probe inserted into the poplar spears with paraffin section method,to do microscopic observations and further study.The results showed that: the roughness of microdialysis probe was about 20 times greater than the diameter of poplar cells;probe caused a certain degree of damage to the surrounding cells;so the collecting dialysate would be contaminated by damaged cell fluid,after probe inserting poplar spears in a period of time,with the time passing ofmicrodialysis,dialysate was close to apoplastic sap.This study explained that the microdialysis technique could get the plant apoplast sap from the micro-structural changes of cells,providing a cytological basis for the application of the microdialysis technique in the plant apoplast.
