Expression and its significance of tumor susceptibility gene 101(TSG101) in gastric cancer cell lines

Objective To study the expression and significance of tumor susceptibility gene 101(TSG101) in different gastric cancer cell lines. Methods The expression levels of TSG101 mRNA and protein were determined using nested RT-PCR、DNA recombinant technology and western blot in gastric cancer cell lines. Results The full-length TSG101 and 288bp aberrant spliced variants were detected in all gastric cancer cell lines. Mutations of the full-length TSG101 were not found by DNA sequence analysis and its expression level was higher in SGC7901/VCR and SGC7901/ADR than in SGC7901. Another 874bp aberrant splicing variant was detected in gastric cancer cell lines MKN45, SGC7901 and its multidrug resistance sublines SGC7901/VCR, SGC7901/ADR. Conclusion The expressions of the full-length TSG101 and its spliced variants may be common in gastric cancer cell lines. The full-length TSG101 may participate in multidrug resistance of gastric cancer.
