Characterization of mycelia growth and phylogenetic analysis of Lentinula edode based on ITS sequence

Lentinulla edodes, knows as Oak mushroom, has been widely used of nutritional and medicinal purpose. This study was conducted to screen the suitable conditions for mycelial growth and to determine the phylogenetic relationship of 58 L. edodes strains. Optimal mycelial growth was observed at 25°C and minimum mycelial growth observed at 10°C and 30°C on PDA (Potato Dextrose Agar) media. Genetic diversity of L. edodes strains was assessed using ITS sequence analysis. A phylogenetic tree based on the ITS region sequences indicated that collected strains could be classified into three clusters. Sequence analysis of the ITS1-5.8S-ITS2 region of 57 strains of L. edodes demonstrated that the ITS region shares mostly 100.0% sequence homology. Interestingly, the ITS region of KNR3093 shares 96.0% sequence homology. The results showed that phylogenetic relationship among the collected strains of L. edodes is high, but genetic diversity that help to develope new variety is low.
