Study on the protective effect and mechanism of action of tanshinoneIIA against ischemia-reperfusion injury in rats

OBJECTIVE To investigate the protective effect of tanshinoneⅡA against isehemia-reperfusionin injury in rats and its mechanism of action.METHODS A total of 60 rats were randomly divided into sham-operation group,model group,tanshinone ⅡA low dose group,middle dose group and high dose group(n=12).The model rats were reproduced by 2 h occlusion following with 24 h reperfusion.The neuro-ethological scores were carried out at 3 h,12 h and 24 h after surgery;cerebral moisture,cerebral index,superoxide dismutase and malondialdehyde in the brain tissues were measured;expressions of cyclooxygenase-2 and nuclear factor-κB p65 were observed with immunohistochemical method at 24 h after surgery,respectively.RESULTS Compared with model group,the neuro-ethological scores,cerebral moisture,and cerebral index in tanshinone ⅡA group significantly decreased,and compared with sham group,the cerebral moisture and cerebral index in model group increased,there was significant difference(P0.05,P0.01).The activity of superoxide increased by 16.27%,18.28% and 24.48%,the content of malondialdehyde decreased by 22.48%,29.12% and 32.04%,the cyclooxygenase-2 expression decreased by 23.19%,28.11% and 30.59%,the nuclear factor-κB p65 expression decreased by 13.68%,18.83% and 22.25% in tanshinone ⅡA pretreatment group(2 mg·kg-1,4 mg·kg-1 and 6 mg·kg-1),respectively,compared with that of model group(P0.05).CONCLUSION TanshinoneⅡA protected against cerebral isehemia-reperfusionin injury by reducing brain edema and scavenging free radicals and inhibiting cyclooxygenase-2 and nuclear factor-κB p65 expression in rats.
