The investigation on the secretion of TNF-#alpha#、IFN-#gamma# by tumor infiltrating lymphocytes

Tumor infiltrating lymphocytes were extracted from fresh stomach adenocarcinoma tissue,and cultivated for 48 h in lymphocytes culture medium,then its concentrations adjuste to 2×10~(6)/ml with 2 ml for special culture. On 4th day the supernatant of TIL was collected and the concentrations of TIL adjuste again to 2×10~(6)/ml with 2 ml for special culture. So do on the 6th ,8th, 10th, 12th, 14th day. ELISA was performed to detect TNF-#alpha#, IFN-#gamma#. The Concentration of TNF-alpha intends to step-up with the elongation of cultivated time. Data shows the 12th day of cultivated time possibly was the turn point of acute increasing. In generally, IFN-gamma intends to step-up with the elongation of cultivated time, and its dose changel more remarkably after 10-12th day of cultivated time. the concentrations of TNF-#alpha#, IFN-#gamma# turn on almost the same trend to step-up with the elongation of cultivated time, and their concentrations all change more remarkably after 12th day, which imply some degree restore of the activity of TIL. So it is not good to perform adoptive immunotherapy without enough clutivated tim eor before 12th day.
