Otoprotectant Activities Comparison of Shudi and Zexie in Different dose proportion on Gentamycin Induced Ototoxic Organotypic Cultures

Objective:To investigate the protective effects of Shudi (radix rehmanniae preparata)-Zexie (Rhizoma Alismatis) prescriptions in different proportion on gentamycin damaged hearing hair cells. Methods:The model of gentamicin induced ototoxicity in cochlear cultures was copied. Cochlear organotypic cultures of postnatal day 3~5 (P3~P5) mice were treated with gentamycin alone or in combination with eight respective Shudi-Zexie prescriptions in different proportion. The effects of the prescriptions on gentamicin induced hair cell loss were observed. Result:Hair cell counting data of the whole organ of corti showed that,compared with the model group,the combination of Shudi and Zeixe in ratio of 8∶3,8∶8,0∶3 and 0∶8 can decrease the hair cell loss ratio significantly (P0.01),while there showed no obvious difference of the other descriptions.Conclusion:The general influence on the gross cell damage rate of the combination of Shudi and Zexie showed a nonlinear dose-effect relationship. The original recipe (8∶3) and the other prescriptions (8∶8、0∶3、0∶8) beongs to the most optimal proportion scales.
