Influence of S-ademetionine on inflammatory factor after injury induced by ischemia-reperfusion in rat liver

Objective:To investigate the influence of S-ademetionine on inflammatory factor after injury induced by ischemia-reperfusion in rat liver.Methods:The total 72 healthy male Sprague-Dawle rats were randomly divided into three group:Sham operation group(Sham group),Ischemic-reperfusion group(IR group),S-ademetionine group(SAM group),24 rats per group.Rats in sham group were administered with the opening and suture of the abdomen with equal NS by intra-peritoneal injection,rats in ischemic-reperfusion group underwent ischemia-reperfusion with equal NS by intraperitoneal injection,and rats in S-ademetionine group was performed with both10%S-ademetionine and ischemia-reperfusion.All rat ischemia-reperfusion models were established by Pringle's method.1 hour after 20-minute ischemic-reperfusion,intraperitoneal injection was given with the dosage of 1 mL/(kg·d) NS or 100 mg/(kg·d) S-ademetionine.Blood and liver samples were collected and analyzed for the levels of tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β)and interleukin-6(IL-6) and hepatic tissue changes were observed at the 1st,4th,7th,10th day after 20-minute ischemia-reperfusion.Results:The serum and hepatic tissue levels of of TNF-α,IL-1β,IL-6 were significantly lower in SAM group than those in IR group(P0.001) at the 1st,4th,7th,10th day after 20-minute ischemia-reperfusion;inflammatory cell infiltration of hepatic tissue were significantly slighter in SAM group than in IR group.Conclusion:S-ademetionine reduce inflammatory factors and inflammatory cells infiltration obviusly and relieve liver injury.
