Cloning and Sequence Analysis of a Disease Resistance Gene from Wild Soybean Seeds

We have cloned some plant disease-resistant gene sequences in existence with some relatively conserved domains. Conserved motifs in NBS domain have been used to isolate resistance gene analogs from soybean. NBS sequence and resistance gene sequence were used as probes to screen a wild soybean cDNA library, conserved motifs such as the nucleotide-binding site (NBS) were found in many characterized plant disease resistance genes. Resistance genes were used as probes to screen a wild soybean (Glycine soja.) cDNA library. A cDNA KR3-x (XLRR) was obtained and we used rapid amplification of cDNA ends (RACE) method from cDNA library and carried out bioinformatics analysis.
