Summary Purpose The aim of this study was to determine the expression of p53 and prevalence of HPV infection in laryngeal squamous cell carcinoma, and to analyze relationship between HPV infection and P53 expression. Material and Method The immunohistochemistry (IHC) staining was used to detect the HPV prevalence and P53 expression in 130 specimens of laryngeal squamous cell carcinoma. Results P53 nuclear expression was detected in 83 (63.8%) out of 130 patients with laryngeal squamous cell carcinoma (LSCC). HPV was identified in 27.7% using IHC of patients with laryngeal squamous cell carcinoma. There was no statistically significant association between the presence of HPV and the epidemiological and clinicopathological features. There was no statistically significant association between the presence of HPV and overall specific survival or P53 expression and overall specific survival. Statistical analysis revealed significant relationship between p53 expression and tumour N stage (p = 0.02), clinical stage (0.4) and localization (p = 0.01) and proved that p53 expression was frequently higher in patients with supraglottic localization and in patients with stage III + IV. Nuclear expression of p53 was detected in 25 (69.4%) out of 36 HPV positive specimens from patients with LSCC. There was no statistically significant correlation between p53 positive staining and HPV infection ( x 2  = 0.67; p = 0.41) A multivariate analysis of survival showed that p53 expression and HPV infection was not an independent prognostic factor in laryngeal cancer Conclusions In conclusion our observations in the current study suggest that the are no relationship between p53 mutation and HPV infection. P53 expression and or HPV presence are not an independent prognostic factor in patients with LSCC
