Establishment and clinical application of a detection method for the infectious hypodermal and hematopoietic necrosis virus in water

Abstract To better understand the distribution of viruses in water, we established a method of concentrating the infectious hypodermal and hematopoietic necrosis virus (IHHNV) in aquaculture. In our study, tangential flow ultrafiltration was established to concentrate the virus from water samples. The results showed that the best filtration columns for the concentration of IHHNV were microfiltration columns (CFP-2-E-4MA) and ultrafiltration columns (UFP-50-C-4MA). The results showed that the average concentration recovery of IHHNV was 67.11%. The method was then used to detect viruses in aquaculture water samples, water samples carried with imported parent shrimp and the dissolved water of imported frozen shrimp. The results showed that the sensitivity of virus detection in water is 46copies / μL, and our methods can be an important tool for aquatic biological security in shrimp ponds and the risk assessment of water carried with imported live animals.
