Study of astragaloside on infiltration of eosinophils in mouse model of allergic asthma

Objective: To observe the effect of astragaloside IV on infiltration of eosinophils in mouse model of allergic asthma,and to explore its possible mechanism.Methods: Using OVA sensitization and challenge to establish the mouse models of asthma and then treated intraperitoneally with astragaloside before each challenge.Observations of clinical symptoms,pulmonary histopathology and inflammatory scores.The levels of inflammatory cytokines and chemokines were measured by Luminex 200.Realtime-PCR and western blotting were also performed to analyze the effects of astragaloside on chemokine receptors expressions.The expressions of NF-κB p65 protein in the lung of mice were tested by western blotting.Results: Compared to the asthmatic model group,the clinical symptoms of mice treated with astragaloside had been relieved.And the extent of inflammation in the lung and the counts of eosinophil in BALF of mice treated with astragaloside were also reduced.The serum levels of cytokines and eotaxin in the mice of astragaloside groups had been decreased too.The expressions of CCR3 mRNA in the lung of mice treated with astragaloside had been significantly decreased(P0.05),and the expressions of CCR3 and NF-κB p65 proteins had also been decreased.Conclusion: Astragaloside can inhibite the infiltration of eosinophils in the mice of allergic asthma.The effects of astragaloside on the expressions of chemokines,chemokine receptors and some cytokines are involved in this process.
