Therapeutic effect of Sishen Wan on lipid peroxidization injury in rats with experimental colitis treated by different ways of administration

Objective:To evaluate therapeutic effect of Sishen Wan on lipid peroxidization injury in rats with experimental colitis treated by different path administration.Methods:Therapeutic effect of Sishen Wan on UC rats were evaluated by colonic injury observed in macroscopic and microcosmic including morphologic change,pathological change,colonic injury score,and cell cycle of enterocyte analyzed by flow cytometry.Mechanism of Sishen Wan was explored in myeloperoxidase(MPO),malondialdehyde(MDA),glutathion peroxidase(GSH-Px) and superoxide dismutase(SOD).Results:Compared with the model groups,many facets were remarkably decreased including colonic wet weight,colon weight index,colonic injury score,production of MDA,cellar number in G0-G1 in the Sishen Wan enema groups(P0.05 or P0.01),and only the same statistical significance of MDA,MPO and cellar number in G0-G1 were found in oral Sishen Wan groups(P0.05 or P0.01).And activity of SOD was markedly increased in in oral Sishen Wan groups(P0.05 or P0.01).Meanwhile,the phenomenon of ulcer restored and the extent of inflammatory cell infiltration were observed in mucous membrane of colon from rats in Two Sishen Wan groups.Conclusion:Sishen Wan could effectively relieve colonic injury of rats with experimental colitis,which potential path was that Sishen Wan might inhibit lipid peroxidization injury,down-regulate contents of MDA and activity of MPO,up-regulate the level of SOD,improve colonic cytothesis to protect damaged colonic mucosa.And effect of Sishen Wan treated rats with experimental colitis by enema outweighed by intragastric administration.
