Surface Enhanced Raman Scattering (SERS) Detection of DNA with Modified Nanoparticles

Surface enhanced Raman scattering (SERS) is a promising tool that provides finger-print spectra for analytes. We introduce modified gold coated magnetic iron oxide (Fe3O4) nanoparticles (MNP@Au) and Raman reporter modified silver (Ag) nanoparticles as SERS substrates to detect target DNA in solution rapidly. Rationally designed probe DNAs were conjugated on the modified nanoparticles separately. With the target DNA presence in the system, the strand acted as a bridge to connect MNP@Au and Ag nanoparticles. Hot spots were formed between two nanoparticles and the signal of the Raman reporter was enhanced. The intensity of the signal is proportional to the concentration of target DNA. This method allows rapid detection of DNA in solution with high sensitivity.
