Protective effects of buddleja officinali flavone on dry eye syndrome and corneal and lacrimal gland in castrated rats

OBJECTIVE To assess the relationships between the protective effect and tissue injury in cornea and lacrimal gland of castrated dry eye rat models by intervention of buddlija officnali flavone.METHODS One hundred and fifty healthy Wistar rats were randomly divided into 5 groups each with 30 rats:the normal group(A),the false operation group(B),the castrated group(C),the therapy group treated with testosterone(D) and the therapy group with buddleja officnalis flavone(E).Bilateral testis and epididymis of group C,D and E were resected;group B were performed scrotal incision only without removing the testicles,as sham-operated control group.Nothing was performed on group A.When the wounds were basically healing,the rats were treated with medicine 1 week after the model was established.Group A,B and C were administered saline one time a day;Group D were treated with testos-terone propionate by thigh intramuscular injection once per 3 days;Group E were treated with buddleja officnalis for one time a day.Ten experimental animals in each group were randomly chosen 1 month,3,5 months after medication respectively,which were performed Schirmer I test,tear film break-up time and corneal fluorescein staining.And then the animals were killed.The protective effect in cornea and lacrimal glands were observed by light microscopy and TEM observation.RESULTS The cornea fluorescence staining:Group A,B had not obviously changed;Group C showed positive after 3 months and aggravate as time;Group D,E the cornea fluorescence were usually dyed a bit and reduced after 3,5 months.SIT:Group C(except group C1) the wet paper was shortened,the difference was significant with before the castration(P0.05),there was no significance with the extension difference of time(P0.05).Group D(except group D1) compared with C,the difference was significant(P0.05).Group E(except group D1) compared with C,the difference was significant(P0.05).BUT:Group C(except group C1) breaks time was shortened,the difference was significant with before the castration(P0.05),and there was significance with the extension difference of time(P0.05).Group D(except group D1) compared with C,the difference was significant(P0.05).Group E(except group D1) compared with C,the difference was significant(P0.05).The light and electric mirror result showed:the texture of lacrimal gland arrang in regularity,the degenerate and necrosising part were deduced,the inflammation of cells was deduced,and then disappeared.But the effect came to weak with extension of the course of disease and the difference was significant compared with the Androgen.CONCLUSIONS Buddleja officnalis flavonoids not only can reduce the gradually increased damage of total male castration of the lacrimal gland and corneal epithelium,but also can mitigate the loss of local inflammation.Dry eye model of the cornea and lacrimal gland has a protective effect.
