Renal effects of streptozocin: Preliminary results of the STREPTOTOX prospective study.

2014
e15155 Background: Based on old studies, streptozocin (STZ), used to treat gastro-entero-pancreatic neuroendocrine tumors (GEP-NETs), is known to have some renal toxicity. We investigated the renal effects of STZ with current dosing regimens and renal surveillance in a 2-part study (retrospective and prospective part). Methods: Patients with GEP-NETS treated with STZ were included. The primary endpoint was the percentage of patients who had a decrease of estimated Glomerular Filtration Rate (eGFR) of at least 25% (MDRD formula) compared to baseline. We report here the results of the prospective part of the study. Results: 30 patients were included and evaluable (median age 64.5 [32-81] yrs, 50% men, mean eGFR 93.2 ± 23.5 ml/min, all patients had well-differenciated tumours and 87% were metastatic. 27 patients received IV STZ only, 3 intra-arterial STZ (chemoembolisation). Median number of cycles was 3 (1-12) for IV STZ. Mean eGFR remained roughly stable during treatment. 4 (15.4%) had eGFR decrease ≥ 25% ...
