MicroRNA- target gene and microRNA- small molecule interaction network analysis of fibroblast like synoviocytes in rheumatoid arthritis

Objective To establish Rheumatoid Arthritis Fibroblast Like Synoviocytes(RA-FLS) background specific microRNA-target gene and microRNA-small molecule interaction network. Methods microRNA PCR Array data(GSE72564) was used to detect differential expression of microRNA between RA-FLS and osteoarthritis FLS.MicroRNA-target gene interaction data(miRTarBase, TarBase, miRecords, TargetScan) were integrated to construct the regulatory networks between the specific expression of microRNA and the target gene associated with RA-FLS.Then, microRNA-small molecule interaction data(SM2miR, Pharmaco-miR) were integrated to construct microRNA-small molecule regulatory network, and network centrality was calculated and core regulatory microRNA and small molecule nodes were obtained. Results 63 differentially expressed microRNA were got and RA-FLS background specific microRNA-target gene and microRNA-small molecule interaction network were established, then five key microRNA(hsa-let-7b-5p, hsa-mir-15b-5p, hsa-mir-218-5p, hsa-mir-30a-5p, hsa-mir-520d-3p) and four active small molecules (5-fluorouracil, glucose, 1, 2, 6-Tri-O-galloyl-beta-D-glucopyranose, glucocorticoid) were screened. Conclusions RA-FLS background specific microRNA-target gene and microRNA-small molecule interaction network, especially key microRNA and small molecule screening, is expected to become a potential target for RA-FLS research.
