Effect of Dai Huang Fu Zi Tang on mitochondrial structure and function of intestinal epithelial cells in rats with severe actue pancreatitis

Objective To investigate the effects of Dai Huang Fu Zi Tang(DHFZT) on mitochondrial structure and function of intestinal epithelial cells (IEC) in rats with severe acute pancreatitis(SAP) . Methods sixty SD rats were randomly divided into three groups: sham group, model group and DHFZT treatment group, each group was then divided into 6, 12, 24, 48 h hour-subsets (n=5). Model and DHFZT group made a SAP model by injecting 5% sodium taurocholate (0.1 mL/100 g) in pancreatic bile duct. DHFZT(10 mL/kg) was injected into rat colon by coloclysis at 0, 15, 30, 45 h post-operation in DHFZT group. The pancreas in sham group was overtured several times, and injected with 0.9% saline at the same time point. To test serum amylase and endotoxins and mitochondrial ATP synthase and cytochrome C oxidase of IEC. Observe intestinal epithelial damage index under light microscope. Results Compared with sham group, level of serum amylase, endotoxin were significantly increased at 6, 12, 24 and 48 h in SAP group. DHFZT could lowered level of them 24 h and 48 h after SAP. Mitochondrial ATP synthase and cytochrome C oxidase activity of IEC were gradually decreased at 6, 12, 24 and 48 h in SAP group comparison with sham group. After treatment with DHFZT, the level of mitochondrial ATP synthase and cytochrome C oxidase of IEC were obviously enhanced at 12, 24, 48 h after SAP. In addition, DHFZT could reduce the intestinal epithelium damage index at 48 h after SAP. Conclusion DHFZT could enhance mitochondrial ATP synthase and cytochrome C oxidase activity of IEC after SAP, and improve mitochondrial energy metabolism and respiratory function, reduce serum endotoxin and alleviate intestinal mucosal epithelial injury.
