Afatinib, an irreversible blocker of the ErbB family of proteins (epidermal growth factor receptor [EGFR], human epidermal growth factor [HER]-2, HER-3 and HER-4), is used to treat advanced EGFR mutation-positive, non-small-cell lung cancer.
