The present invention is a method of synthesizing single-stranded DNA library based on the evolution of metabolic pathways, it belongs to the technical field of genetic engineering. The present invention employs a single-stranded synthetic DNA library, the PCR-mediated technique of recombinant mutant library construction. The recombinant expression vectors after mutation a library of transformed host evolution high throughput screening mutants. The present invention employs this technique, successful transformation of the metabolic pathways in vitro, very excellent properties obtained mutants. The present method enables rapid evolution of genes in vitro, simple operation, especially suitable for introducing the rich diversity internal gene mutations, rapid directed evolution.
