Risk of uterine cancer in BRCA1 and BRCA2 mutation carriers.

1529Background: The risk of uterine cancer (UC) in BRCA1 or BRCA2 mutation carriers is unclear. Previous studies have generally suggested about a 2-fold increase in risk, with one recent study suggesting a much higher relative risk when analysis was limited to serous UC. We sought to assess whether Australian mutation carriers are at increased risk of developing UC compared to the general population. Methods: Prospective cohort study of Australian BRCA1 and BRCA2 mutation carriers in the kConFab familial breast cancer cohort. Eligible mutation carriers had a uterus present and no previous history of UC at cohort entry. Information on cancer events, epidemiological and lifestyle factors was collected by interview at cohort entry and then every 3 years using a mailed questionnaire. Self-reported cancer events were verified with pathology reports obtained from the treating institutions, and date of surgery and histologic subtype abstracted.
