Ultrasonic Extraction of Flavonoids from Herb of Sievers Wormwood and its Optimization by Response Surface Method

In this study, the response surface method on the base of single factor test was used to optimize the extract process of ultrasonic-assisted cellulase extraction of flavonoids from Luliang herb of sievers wormwood. After optimizing cellulase factors by the orthogonal experiment, the ultrasonic factors were studied with Box-Behnken design and polynomial quadratic equation predicting by Expert 7.0 analysis. The best solution was as follows: ultrasonic power 396 W, ultrasonic time 14 min, extracting 3times, 2.5% adding amount of enzyme, enzymolysis time of 120 min at 50 ℃ and p H 5.5. Under the optimized condition, the predicted and actual extraction rate of flavonoids were respectively 2.43% and 2.47%. Thereby the fitted multiple regression equation was credible.
