Association between genetic polymorphisms of ADAM33 with asthma and its phenotypes

Background: ADAM33 is one of the positionally cloned asthma associated genes, which has been identified as susceptibility gene for asthma in different populations. Aims and Objectives: To study ADAM33 polymorphisms and understand their role in asthma phenotypes, asthma severity and response to treatment after 2 months of ICS and LABA in south Indian population. Methods: A total of 989 individuals including 503 asthma cases and 486 normal controls were genotyped for 6 ADAM33 SNPs using MassARRAY technique with a MALDI-TOF MS analyzer. SNPs of ADAM33 were correlated with various phenotypes of asthma. Haplotype analysis using Haploview software V.4.0 was done to assess the association between haplotype of ADAM33 SNPs and asthma. Results: There was no association of any ADAM 33 SNP with asthma. There was no association with either atopic or non-atopic asthma. On subgroup analysis, rs2853209 an association with TT genotype and minor T allele was observed with moderate asthma (P Conclusion: We observed association of rs2853209 with moderate asthma and rs2787094 with FEV1 and FVC reversibility after treatment and two marker haplotype associations with asthma in south Indian population. Further in-vitro functional studies are required to confirm the associations.
