Low-dose heparin on the role of early intervention in septic rats

Objective To explore the early intervention role of the application of low-dose heparin on sepsis through rats in cecal ligation and puncture(CLP) model.Methods 72 Wistar rats were randomly divided into three groups,namely sham operation group(sham),CLP group(CLP) and heparin group(heparin),n=24.Each group was divided into three sub-groups by 6 h,12 h,24 h three time points,each sub-group 8,respectively,the three time points of various sub-routine testing for blood in rats in rat blood platelets,APTT,with the ELISA assay in plasma TNF-α content.The specimens from lung tissue were HE staining to observe pathological changes in lung tissue.Results Compared with CLP group and sham group,platelets in the 12 h and 24 h significantly reduced(P 0.01),heparin group at 24 h also reduced(P0.01),but higher than that of the CLP group(P0.01),APTT at 24 h,CLP Group was significantly longer than that of the sham group(P0.01),compared with sham group,heparin group also extended(P0.01),but shorter than that of CLP group(P0.01).TNF-α levels in plasma at different time points CLP group,heparin group were higher than that of the sham group(P0.01),but the heparin group at 12 h and 24 h were lower than that of the CLP group(P0.01).Compared with CLP group,the level of congestion and inflammatory cell infiltration was significantly ameliorated in heparin group in light microscopic observation of lung tissue.Conclusion Heparin might reduce blood levels of TNF-α through the intervention in coagulation-inflammation network of sepsis,thereby it can also reduce inflammation in lung tissue damage,play a role of intervention in early alleviate the occurrence and development of sepsis.
