Assessment of effect of olmesartan medoxomil on endothelial dilation function in patients with isolated systolic hypertension

2012
OBJECTIVE To observe the effect of olmesartan medoxomil on endothelial dilation functionin patients with isolated systolic hypertension(ISH).METHODS Fourty patients of ISH were randomized to receive olmesartan medoxomil(treatment group) for 12 weeks. The baseline of biachial angiobore and changes of it at respone to nitroglycerin and reactive hyperaemia were assessed by high frequency ultrasound before and after treatment, and were compared with those in 40 healthy elderly people(control group).RESULTS After treatment of olmesartan medoxomil for 4 weeks the levels of SBP、DBP and PP were significantly reduced(P0.01 or P0.05 respectively). After 12 weeks of treatment the levels of SBP and PP were significantly lower(P0.01) than those after 4 weeks, but the decrease of DBP was not statistically significant(P0.05).Before treatment the brachial endothelial-dependent vasodialation during reactive hyperemia(flow-mediated dilation,FMD) was reduced(P0.01) in treatment group than that of control group. After treatment of olmesartan medoxomil for 12 weeks the FMD was improved(P0.01).There were no significantly change of vasodilation in response to nitroglycerin(NMD) before and after treatment(P0.05 all).No severs adverse effect was found in treating with olmesartan medoxomil.CONCLUSION Olmesartan medoxomil has good curative effect and is safety on ISH,and it has improvement action of FMD.
