The effect of new therapy of 20 AA compound amino acid plus high dose of vitamin B6 on the expression of coagulation factor genes in rat model of trauma-induced coagulopathy

Objective To investigate the effect of new therapy of 20 AA compound amino acid injection plus high-dose vitamin B6 on the gene expression of coagulation factors in liver of rat with trauma induced coagulophathy(TIC) model. Methods Thirty male SD rats were randomly(random number) divided into normal group, sham group, control group and new therapy group, and then control group and new therapy group were divided into four sub-groups (4 h, 6 h, 12 h and 24 h after injury, n=3 in each group). The multiple trauma model of rat was established by using Feeney's free fall epidural impact method, plus bilateral femoral fractures and bleeding. According to above intervals, the total RNA was isolated from liver tissue by Trizol method, the mRNA expression level of coagulation factor genes (F Ⅱ, F Ⅶ, FⅨ, F Ⅹ, F Ⅺ, F Ⅻ, F ⅩⅢ A subunit) were determined by real-time PCR method. Results Compared with normal group, except for F Ⅺ and ⅩⅢ A subunit, the expressions of coagulation factor genes in control group and new therapy group were up-regulated significantly from 4 h after injury, but the expressions were down-regulated with time prolonged. Compared with control group, the expression was higher in the new therapy group(P<0.05). Conclusions The new therapy can improve the coagulation function by enhancing the expression of the coagulation factor genes in the liver.
