Experimental study of the interference of Hongjingtian in cytokines in hippocampus of post stroke depression rats.

Objective To study the interference of Hongjingtian in cytokines in hippocampus of post stroke depression(PSD) rats.Methods The healthy SD rats were randomly divided into five groups:simple focal cerebral ischemia group,PSD group,Hongjingtian group of three doses [2.5,5,7.5 g/(kg·d)].PSD rats model was established.Hongjingtian group was given Hongjingtian and other groups were given normal saline.The ethological score of depression was evaluated.Pathological changes of hippocampus were observed by HE staining and the content of TNF-α,IL-1β and IL-6 in hippocampus were measured.Results Compared with normal group,the expression of the content of TNF-α and IL-1β of other groups increased,while the content of IL-6 decreased,the ethological score of depression decreased.HE staining of hippocampus showed neurocyte damaged obviously.The difference between them was significant(P 0.01).Compared with simple focal cerebral ischemia group,the content of TNF-α and IL-1β in model group increased,while the content of IL-6 decreased.The ethological score of depression decreased.HE staining of hippocampus showed neurocyte damaged obviously.The difference between the two groups was significant(P0.01).Compared with model group,Hongjingtian groups decreased the content of TNF-α and IL-1β,increased the content of IL-6,and increased the ethological score of depression,reduced the damaged extent of neurocyte of hippocampus.The difference between the two groups was significant(P 0.01).The effects of medium-dose and high-dose groups were more obviously compared with the low-dose groups(P 0.05),but there was no difference between them(P 0.05).Conclusion The steady state of network of cytokines in PSD experienced disequilibrium and neurocyte of hippocampus were damaged worse than that in simple focal cerebral ischemia.Hongjingtian has neuroprotective effects on PSD rats,its mechanism may through improving the damaged extent of neurocyte of hippocampus by way of reducing the content of TNF-α,IL-1β and increasing the content of IL-6 in hippocampus.
