Abstract 16956: The Therapeutic Potential of Human iPS Cell-derived Cardiac Tissue onto a Porcine Myocardial Infarction Model

Introduction: We previously generated human induced pluripotent stem cell (hiPSC)-derived cardiac tissue sheets (CTSs) including cardiomyocytes, endothelial cells and vascular mural cells, which improved cardiac function after myocardial infarction. We also reported that thick layered CTSs by the insertion of gelatin hydrogel microspheres (GHMs) between each layer allowed to engraft as multiple layered cardiovascular cells accompanied by functional capillary networks in a rat myocardial infarction (MI) model (human iPSC-derived cardiac tissue: HiCT). Here we aimed to validate the therapeutic effect of 5-CTS layered clinical-scale large HiCT (HiCT-5L) in a porcine MI model. Methods: HiCT-5L were generated by inserting GHMs to the space between CTSs (3.5cm in diameter). Female mini-pigs were divided into two groups: HiCT-5L treatment (T) and sham (S).
