THE POTENTIAL OF PHARMACODYNAMIC INTERACTION OF GARLIC AND METOPROLOL IN RATS WITH MYOCARDIAL DYSFUNCTION AND METABOLIC DERANGMENTS

The present study was aimed to determine the therapeutic efficacy of metoprolol in presence and absence of aqueous extract of garlic in rats with Isoprenaline induced myocardial damage. Sprague dawley rats were treated with GH (250 or 500 mg/kg, p.o.) for three weeks. In the interactive groups, metoprolol was incorporated in the garlic aqueous extract therapy in the third week. Isoprenaline (ISO, 150 mg/kg, sc) was injected at the end of treatment period. Blood samples were collected to serum evaluation of LDH and CK-MB. Hearts were removed from anaesthetized rats and heart tissue homogenate was prepared in 0.25 M sucrose for evaluation of SOD Catalase and TBARS. Hearts were subjected for histopathological studies. In serum there was significant reduction in LDH, CKMB in GLD, GHD, metoprolol, GLD+ metoprolol and GHD+ metoprolol when compare to Isoprenaline control. In HTH there was significant increase activity of LDH, in groups treated with, GLD, GHD, metoprolol, GLD+ metoprolol and GHD+ metoprolol and with CKMB activity increased in GLD, metoprolol and GLD + metoprolol. Because of pretreatment with GLD, GHD, metoprolol, GLD+ metoprolol and GHD+ metoprolol there were increased SOD Catalase activity as compare to ISO group in HTH. TBARS activity increase in ISO group as compare to normal group in HTH, but there was decrease TBARS level in groups treated with GHD, metoprolol, GLD + metoprolol GHD+ metoprolol. From the present results it may be concluded that the GLD (250 mg/kg) and GHD (500 mg/kg) both possess cardioprotective efficacy when given to Isoprenaline induced M.I. in experimental animals. Moreover, combined therapy of GLD and GHD and metoprolol demonstrated synergistic potential cardioprotective activity.
